چکیده

The calcium-triggered photoprotein aequorin has been used as a sensitive indicator of cytoplasmic free calcium in cells of relatively large size. We describe here several novel modifications to the technique that make it possible to detect resting levels of free calcium in single mammalian oocytes, fibroblasts and granulosa cells. Concentrations of free calcium as low as 1 X 10(-7) M have been measured in single fibroblasts, corresponding to a detection limit of 10(-18)mol.

Utilizing Hidex Sense for mitochondrial and cytosolic calcium measurements using the luminescent calcium reporter protein aequorin and the fluorescent calcium indicator Fura-2 AM

reporter. Aequorin is a Ca 2+ -sensitive luminescent protein originally derived from the jellyfish Aequorea victoria. When expressed in mammalian cells, wild-type aequorin localizes to the cytoplasm and can be employed for cytoplasmic Ca 2+ measurements. Recombinant aequorin fusion proteins with specific targeting peptide sequences have been produced in order to direct the reporter protein to a...

Different Stress-Induced Calcium Signatures Are Reported by Aequorin-Mediated Calcium Measurements in Living Cells of Aspergillus fumigatus

Aspergillus fumigatus is an inhaled fungal pathogen of human lungs, the developmental growth of which is reliant upon Ca2+-mediated signalling. Ca2+ signalling has regulatory significance in all eukaryotic cells but how A. fumigatus uses intracellular Ca2+ signals to respond to stresses imposed by the mammalian lung is poorly understood. In this work, A. fumigatus strains derived from the clini...

Ca2+ Measurements in Mammalian Cells with Aequorin-based Probes.

Aequorin is a Ca2+ sensitive photoprotein suitable to measure intracellular Ca2+ transients in mammalian cells. Thanks to recombinant cDNAs expression, aequorin can be specifically targeted to various subcellular compartments, thus allowing an accurate measurement of Ca2+ uptake and release of different intracellular organelles. Here, we describe how to use this probe to measure cytosolic Ca2+ ...

Cellular calcium fluctuations in mammalian heart: direct evidence from noise analysis of aequorin signals in Purkinje fibers.

Indirect evidence suggests that fluctuations, or oscillations, in the intracellular free calcium concentration [( Ca2+]i) can occur spontaneously in intact cardiac preparations, but such [Ca2+]i fluctuations have never been demonstrated directly. We used the bioluminescent Ca2+-sensitive protein aequorin to detect fluctuations in the [Ca2+]i in canine cardiac Purkinje fibers. Noise analysis of ...

Measuring [Ca2+] in the endoplasmic reticulum with aequorin.

The photoprotein aequorin was the first probe used to measure specifically the [Ca(2+)] inside the lumen of the endoplasmic reticulum ([Ca(2+)](ER)) of intact cells and it provides values for the steady-state [Ca(2+)](ER), around 500 microM, that closely match those obtained now by other procedures. Aequorin-based methods to measure [Ca(2+)](ER) offer several advantages: (i) targeting of the pr...
